Other big stories in the FA Cup include League Two Grimsby Town beating Premier League Southampton to set up a quarter-final with Brighton and Hove Albion; while Championship leaders Burnley travel to Manchester City.
Manchester United, who knocked Reading out in the fourth round, host Fulham in the other quarter-final.
Reading beat fourth-placed Rovers in the reverse fixture 3-0, their first defeat of the season.
