The Championship outfit were no match for Mikele Leigertwood's Young Royals, with midfielder Kiyan Coke-Miles-Smith scoring a hattrick at the famous old ground.
Taylan Harris, who scored in the EFL Trophy on debut in the historic 9-0 win over Exeter City, added extra gloss to the comfortable win.
A date for the draw is yet to be confirmed, with last night's match the first of 16 ties to take place.
It came on the same night as the first team's EFL Trophy exit to Brighton and Hove Albion Under-21s at the Amex Stadium.
Reading's best achievement in the historic tournament is a semi-final appearance in 2015, losing to Fulham over two legs under the management of David Dodds.
