Reading, Cheltenham Town and Newport County will be joined by West Ham United Under-21s in their Bristol Street Motors Trophy group.
A minor competition to allow lower league teams the opportunity to play at Wembley Stadium, the controversial decision to allow youth teams in the tournament came eight years ago.
Split between north and south, exact fixtures and dates remain unknown.
All youth teams have to play away from home in the group, with Reading guaranteed one away match.
Last year, the Royals cruised through the group stage without dropping a point. Eventually, they fell at the round of 16 stage when Brighton and Hove Albion Under-21s knocked Ruben Selles' team out.
West Ham fell at the same hurdle last year, missing out to eventual finalists Wycombe Wanderers.
This draw came just hours after the EFL Cup draw was made, pitting Reading against Colchester United in round one.
