Reading are among the oldest professional sides not to have won the prestigious competition, coming closest in 1927 and 2015 with semi-final appearances.
The quarter-final was also reached in 2010, 2011 and 2016.
Last season, the Royals made it to round two before losing to National League Eastleigh live on television, a game best remembered for fan protests against owner Dai Yongge.
Premier League and Championship teams enter at the third round stage.
