The Bees, of Southern League Premier Division Central, knocked out Tonbridge Angels in round one.
Last season, MK Dons were defeated in round one before National League Eastleigh caused a cup upset and knocked Ruben Selles' side out at the Silverlake Stadium, similar to 2021 and an infamous defeat at Kidderminster Harriers.
Fixtures are due to take place over the weekend of November 29-December 2, with details to be confirmed once television companies choose which matches are to be televised.
Premier League and Championship sides enter at round three, which comes on the first weekend of January.
Reading reached the semi-final in 2015 and are among the oldest professional clubs to never reach the showpiece final since its foundation in 1871.
